Test Principle:It employs a substrate Ac-IETD-pNA (acetyl-Ile-Glu-Thr-Asp p-nitroanilide) that can be cleaved by caspase-8 into a yellow chromophore p-nitroaniline (pNA) product. The amount of free pNA produced is proportional to the enzymatic activity of caspase-8 and can be quantified using a spectrophotometer or a microplate reader at 45nm.
