Test Principle:After PI staining, If the fluorescence intensity of cells in G0/G1 phase is consider to be 1, the theoretical value of the fluorescence intensity of G2/M cells containing two copies of genomic DNA is 2 and the fluorescence intensity of S-phase cells undergoing DNA replication is between 1-2. Due to the condensation of nucleus and DNA fragmentation in apoptotic cells, some genomic DNA fragments are lost during the PI staining. Therefore, apoptotic cells exhibit weak PI staining with a fluorescence intensity less than 1, and a sub-G1 peak, also named as apoptotic cell peak, appears when examined by flow cytometry.
