Test Principle:JC-1 is a cationic dye that exhibits potential-dependent accumulation in mitochondria.  In normal cells with higher mitochondrial membrane potential, JC-1 aggregates in the matrix of mitochondria and forms the red fluorescent J-aggregates. However, in cells with lower mitochondrial membrane potential, JC-1 does not aggregate and exists as green fluorescent monomer. Therefore, the changes of mitochondrial membrane potential in cells can be conveniently determined by examining the shifting of fluorescence, and mitochondrial depolarization can be indicated by a decrease in the red/green fluorescence intensity ratio.
