Test Principle:Mitochondria in normal cells sequester a large amount of positively charged TMRE due to the negative charge inside mitochondria and emits bright red-orange fluorescence. However in apoptotic cells. the intensity of red-orange fluorescence inside mitochondria decreases significantly because the mitochondrial membrane potential is lost and TMRE is released into the cytoplasm through the opened mitochondrial permeability transition pore (MPTP). Therefore, the changes of mitochondrial membrane potential during the onset of apoptosis or necrosis can be determined by examining the fluorescence intensity in mitochondria of cell samples.
