Test Principle:Beyotime T4 Endonuclease V, also known as T4 Pyrimidine Dimer Glycosylase (T4 PDG) is a recombinant enzyme derived from the T4 phage and serves as a DNA damage repair enzyme with both DNA N-glycosylase and AP-lyase activities. Its N-glycosylase activity recognizes cis-syn-cyclobutane pyrimidine dimers (T^T, T^C, C^C) caused by UV irradiation and cleaves the 5'-glycosidic bond, generating an Apurinic/Apyrimidinic (AP) site. Its AP-lyase activity then removes the AP site via β-elimination, creating a one nucleotide DNA gap with 3'-α,β-unsaturated aldehyde and 5'-phosphate termini.
